Any ideas which ports Lufthansa will fly their first A380 between? Also will Air France expand to different ports with their second delivery?

I heard that Lufthansa's first A380 route will be FRA-JFK and that Air France's second A380 route will be CDG-JNB.

Yes that is right Air France flights to JNB start on 17th Feb.

Lufthansa's third A380 (MSN044) will hopefully make its first flight tomorrow.

Lufthanasa's 3rd A380 (MSN044) performed a rejected take off test on Friday. But first flight was cancelled due to bad weather. First flight is now scheduled for Monday.
